Manifold Pressure

When my plane is not running the manifold pressure guage reads 1" below ambient pressure. I do not worry about this as it's very consistant.

My question is this. If I want to set 75% power by the POH, would I need to reduce the manifold pressure 1"?

Yes, assuming that your error is constant throughout the entire range.

If you are based at an airport with a field elevation of 1,000 feet (+/-) then your MP gauge is fine. If you are at sea level, then adjust as Zach suggests.
